Found at postsynaptic densities of excitatory neurons, PSD-95 plays an important role in synaptic plasticity and the stabilization of synaptic changes during long-term potentiation.

Encoded by DLG4, Postsynaptic Density Protein-95 (PSD-95) functions as a major scaffold protein that can cluster membrane receptors, ion channels, including NMDA receptor subunits, Kv1 potassium channels and Caspr2. PSD-95 has the unique ability to interact with a large number of proteins through three PDZ domains, followed by SH3 and GK domains. PSD-95 is highly regulated by phosphorylation. PSD-95 disruption has been associated with cognitive and learning deficits such as Alzheimer’s, depression, schizophrenia, and autism.
