"PBMCs were seeded in a 96-well plate at a density of 1.6 × 105 cells/well (8.0 × 105 cells/mL). Relevant wells were primed with 100 ng/mL LPS for 4 hours and, if required, then stimulated with 10 mmol/L ATP for 30 minutes. Caspase-1 activity was measured by using FLICA (ImmunoChemistry Technologies, Bloomington, Minn), a cell-permeable fluorescent probe (FAM-YVAD-FMK) that binds active caspase-1.
