Recombinant Antibody Development
Antibody discovery (custom antibody development) continues to be done primarily in animals (mice, rabbits, chickens ...) due to the incredible power of the immune system to select and affinity mature antibodies in vivo. While "panning" of synthetic antibody libraries has gained popularity, these libraries cannot match the diversity of antibodies developed in vivo. However, once discovered, there are many advantages to producing monoclonal antibodies in a recombinant format.
Expression Optimization
Mouse monoclonal hybridoma cell lines often produce antibody at low levels. Recombinant antibody expression platforms create opportunities for optimization at each step through:
- codon optimization to ensure maximum antibody production from a known source
- use of a highly productive promoter and secretion signal
- highly optimized growth conditions - since recombinant antibody production is routinely performed in just two different cell lines (HEK293 and CHO), production conditions have been highly optimized for these cell lines enabling high-level antibody production and reproducibility.
Antibody Engineering
Once a monoclonal antibody has been sequenced, it can be engineered into a growing number of formats.
- Monoclonal antibodies can be reformatted with the conserved Fc domain from another host creating a "chimeric" antibody.
- Antibody sequences from two different antibodies can be engineered into a single protein creating a "bispecific" antibody
- Antibody effector function, stability, and other attributes can all be manipulated.

Hybridoma Sequencing
Determining the sequence of the variable domains of the antibody heavy and light chains is the first step towards creating a recombinant antibody. Hybridoma sequencing also ensures that you never lose your precious monoclonal antibodies or continue to pay for their storage.
Expression Plasmid Cloning
- We use the sequence you obtained from hybridoma sequencing (or provided directly by you) to create your antibody expression plasmid.
- We use our proprietary expression plasmids with optimized promoter and secretion signal to ensure maximal yield.
- Deliverables include 10 ug of expression plasmid and plasmid map. Optional deliverables include glycerol stock and plasmid scale-up.
Antibody Expression and Purification
- Suspension-adapted HEK293 cells are transiently transfected to induce high yield production of antibody.
- Protein A purification allows for purification of most IgGs. Custom affinity purification is available for atypical formats (please inquire).
- Antibody purity >95%
