Plasma Sample Diluent should be utilized as a buffer matrix for diluting out plasma samples in antigen-down or antigen-capture ELISA formats. This diluent is also an excellent choice for diluting out antigen standards in ELISA standard curve preparation. This ELISA sample diluent contains proprietary additives in a buffered BSA protein base providing an assay environment that minimizes nonspecific binding while maintaining proper ionic strength and pH for efficient antibody-antigen interaction.
Specially formulated additives inhibit thrombin activity (clotting) and complement activity during the plasma sample incubation period. Plasma Sample Diluent also contains antimicrobial agents to allow for room temperature benchtop use and extensive storage stability.

When diluting test samples, first note the dilution factor for future data analysis. For example, a human plasma sample was diluted 1:10 (50 µL plasma sample + 450 µL plasma sample diluent) and yielded a standard curve derived analyte concentration of 50 ng/mL from the ELISA assay. This concentration should then be multiplied by 10 (the dilution factor) to obtain the actual concentration value of 500 ng/mL in the undiluted plasma sample.
