When diluting test samples, first note the dilution factor for future data analysis. For example, a human plasma sample was diluted 1:10 (50 µL plasma sample + 450 µL plasma sample diluent) and yielded a standard curve derived analyte concentration of 50 ng/mL from the ELISA assay. This concentration should then be multiplied by 10 (the dilution factor) to obtain the actual concentration value of 500 ng/mL in the undiluted plasma sample.
