Anti-Metabotropic Glutamate Receptor Type 2 / Type 3 (mGluR2/mGluR3) Antibody
We have three different anti-mGluR antibodies, each of which binds to a specific peptide sequence(s) present in different sets of mGluR gene family members. The ER2/3 antibody represents sequences found in both mGluR2 and mGluR3. (Note that this product cannot distinguish between these two receptors.) It comprises two antipeptide antibodies generated against overlapping sequences specific to the mGluR2 and mGluR3 gene products but not found in other mGluR gene family members. This antibody has been validated for immunohistochemistry and immunocytochemistry using mouse brain sections and cultured neuroblastoma cells as well as in rat brain sections.

The metabotropic glutamate receptors (mGluRs) represent a gene family of G-protein-coupled receptors, all of which bind the amino acid glutamate (the major excitatory neurotransmitter of the nervous system) but differ in their primary amino acid structure, their pharmacology, and their distribution in the peripheral and central nervous systems. In general, mGluR1 and mGluR5 receptors are post-synaptic and mGluR2 and mGluR3 receptors are pre-synaptic.

Store at 4°C in the dark. Under these conditions, the antibodies should have a shelf life of at least twelve months, provided they remain sterile. For longer term storage, aliquot and freeze to avoid freeze-thaw of the antibody.

Chickens were immunized with two synthetic peptide / keyhole limpet hemocyanin (KLH) conjugates. These synthetic peptides corresponded to unique sequences within the metabotropic glutamate receptor type 2 and type 3 gene products, but was shared between the human (Q14416.2, Q14832.2 NCBI) and mouse (Q14B12.2, Q9QYS2.1, NCBI) sequences. After repeated injections, immune eggs were collected, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using a peptide column, the concentrations of the eluates adjusted to 200 µg/mL, and the preparation was filter-sterilized.
Phosphate-buffered (10 mM) isotonic (0.9%, w/v) saline (“PBS," pH 7.2) with sodium azide (0.02%, w/v) added as a preservative.

This anti-peptide antibody preparation was analyzed by immunohistochemistry (at a dilution of 1:2000) using fluorescein-labeled goat anti-chicken IgY (1:500 dilution, Aves Labs Cat.# F-1005) as the secondary reagent.
